If a US citizen marries a Canadian and then he moves to Canada, can he work and what are the stipulations?

It takes 8 months or more for the initial stage of the application to be approved with will include work authorization. When your wife submits the sponsorship application a request for an open work permit should also be submitted with the application. Once you receive the approval and the open work permit you can start working.

(in reply to: Questions as I am confused) the best option in my opinion is to apply for PR, it usually takes ~6 months, after landing you would be a PR and have the right to work.
you can apply for a work permit of course but only if you have a job offer, in this case you can obtain it within 1-2 months, otherwise just wait for you PR application
