I am going to File for PR myself, I am a Indian, lived in Australia and presently in US.
Do I need to get the clearnece before I file or that comes later. If yes What is the procedure to get the clearnece

Get all the Police CC from these countries except from the US as early as possible. Its not mandatory upon submission but will for sure expedite ur background check process once u get there. Apply for a US PCC 6 months after u get ur file #. This would squeeze ur total PR process to 9 months unlike usual 15 months.

If applying to Buffalo as of Oct 2004 they require you to submit clearance from FBI at the time of initial submission of application.
