Do I need to mention brothers and sisters in additional family information form though they are not accompanying with me.
Should this form be filled by both the applicant and the Spouse seperately?

Yes, You should.
And you and your wife should fill this form too..... They need to keep records of your and your´s spouse family tree..in case you file PR for your or yours spouse´s family member...
